The goal | The objective of the course is to highlight the remarkable potential of mass spectrometry (MS) for the identification and quantification . In this context, the combined techniques are particularly emphasized, GC-MS and HPLC-MS, for the analysis of complicated mixtures of organic compounds, as well as ICP-MS methods for elemental and isotopic analyses. | |||
The outcome | During the course, the student should gain knowledge that enables him to apply mass spectrometry in a variety of fields, from structure testing and compound identification to quantitative tracing of analytes in complicated mixtures. He should also develop experimental skills in applying the combined techniques of GC-MS, GC-MS/MS, HPLC-MS, HPLC-MS/MS and ICP-MS. | |||
Contents | ||||
Contents of lectures | The program covers the classical contents, which explain the basic principles of mass spectrometry, but also recent developments, especially in the field of ionization techniques (EI, CI, APCI, ESI, MALDI) and the application of different types of analyzers (magnetic, electrostatic, quadrupole, different types of ionic traps, flight time analyzer). The characteristics of different combinations of analyzers for hybrid instruments are covered, which allow a considerable expansion of the application of mass spectrometry, both as methods of chemical analysis and as a research discipline. Ion fragmentation reactions are used to identify organic compounds, the so-called MS-MS spectra, and the GC-MS or HPLC-MS or MS-MS combined techniques will be used for quantitative analysis of complicated mixtures, as will be demonstrated in the laboratory. A particular attention will be devoted to isotopic analysis and qualitative and quantitative ICP-MS analysis of metals. | |||
Contents of exercises | Laboratory exercises: Obtaining of mass spectra of compounds and isotopic composition of elements, tandem mass spectrometry, gas and liquid chromatography with mass spectrometric detector, inductively coupled plasma with mass spectrometric detector. | |||
